When you first load into the Inverted Spire, you're in a Public Space (much like Savathun's Song or the mid-point of Exodus Crash). While in a public space, you cannot earn points thus having a 0x multiplier. When you load into Tree of Probabilities, however, the game loads you into an isolated instance (because of the Infinite Forest), thus you may earn points immediately.
You'll need to progress a short bit into the Strike to begin earning points.

We started running the inverted spire and when the points started accumulating the points didn’t add up to what we fill they should been.

When we started the nf with a 5.55x multiplier and when we finished the nightfall we only had a score around 155k when I see people soloing the nightfall with the same multiplier and finishing with over 200k score.

How long did the Nightfall take you? Did you get Fallen or Cabal enemies?

We got cabal and we finished right over 15 minutes. I think it was 15:56.

Don't forget that points decrease after 15 minutes. Inverted Spire, only Vex spawns more enemies as well. I would see the video (probably Eso) and check their add types. It sounds like you guys weren't killing every available enemy.

So killing vex where the spire forms will not give us any points?

Correct. Once you get past the second gravity lift and enter that area there (where the Red Legion and Vex fight eachother) you'll start earning points. The reason for this is because if they allowed you to earn points in Public Spaces, much like the Lost Sector glitch, players would be able to wander off to find a Glimmer Extraction to farm points.

so that will not happen in Tree of Probabilities - we start from a public space there too, right? AT least - did not for us when we started it. Also, does not explain why the score multiplier will show 0.0x instead of the nightfall card score multiplier. Perhaps that needs to be corrected so there is no confusion. I just want to make sure that it is indeed 'not a issue' and is working as expected....

There's a difference -- Tree of Probabilities is an "instanced Strike", it loads you into a separate instance meaning no other players external from the Strike are present, thus you may immediately begin earning points -- there are no Public Events available in that space, no external enemies, etc. Inverted Spire is a "non-instanced Strike", it loads you into a patrol instance, meaning other players may be present when you start the Strike, there are Public Events happening, external enemies, etc. thus you may not immediately begin earning points. Savathun's Song is a "non-instanced Strike", it loads you into a patrol instance, meaning other players may be present when you start the Strike, thus you may not immediately begin earning points. Lake of Shadows is "non-instanced" A Garden World is "instanced" The Arms Dealer is "instanced" etc. The multiplier on your score card reflects the multiplier of points at that very moment -- if it loaded you in with a 4.65x multiplier but you're really earning 0x the points, that would cause a lot more confusion that the multiplier changing depending on the circumstances. Once you enter an instanced area, the multiplier will change to 4.65x from 0x, meaning you're now earning 4.65x the points. Once you reach the 15 minute mark, that multiplier now goes down to 2.33x, meaning you're now earning half the points.
