EDIT:Deej most certainly did not mean the Treasure Room in the Prison of Elders of the Tower in the vault as there is no secret to them. because what other vault would have secrets in this game?
EDIT: Most say its a figure of speech but no one can tell me why. mostly cause it's not. a figure of speech is an example not an statement.
Most recently (5-21-2015) Deej had said in his update. here is a quote from that update

And all of it had me thinking again. Why would he randomly mention the Vault of Glass in the House of Wolves update? It's completely irrelevant. Although there is one mission in House of Wolves that takes place in the Templar's room, it still has nothing to do with it. Now break down what Deej stated.

a secret or two? everyone knows the spirit bloom chest isnt considered an actual raid chest since it gives no weapons or armor.
and here's another thought. If you go back to where you fight Zydron, The Gate Lord, You'll notice the enemies aren't in a darness zone for the raid but they're higher level than normal. and if you find Kabr's Ghost his grimoire states

upon further reading it talks about a would as you imagine it, basically describing the Vault of Glass. the gorgons decide if you are real and wipe you from existence if the decide you don't exist.
the crazy parts about all of this
1. Why would Kabr's body be near another vex portal and not inside the vault of glass? [spoiler]it was meant to be found there by bungie[/spoiler]
2. why is the grimoire that's found on his body directly relating between the numbers 5-7(7 is Bungie's favorite)

3. why would Deej bring something like this up 6 months after the Vault had been dominated?[spoiler]They might have added more to the vault but possibly only adding the trigged mechanisms to activate the last 2 chests[/spoiler]
Before anyone flips out saying "It's been six months they would've found it now!" remember that it took someone 7 years to stumble upon an Easter Egg in Halo 3 that was released in 2007!! if it takes 7 more years I'll find the chests. anyone else have anyone speculation?

It's after you kill Atheon I reckon, that's why you don't get bumped to orbit. Go through one of the gates and it's behind a pillar or something.

Here is a completely crazy thought. What if the spirit bloom chest is actually a raid chest but a series of events needs to happen first to make it one? For example, when the Oracles first appear - as in the 'here I am' sequence - you can actually damage the oracles. Perhaps if all 6 guardians stood in 6 of the Oracles positions and guardian 1 moved to Oracle 7 and all guardians killed the oracles it might do something?

I think he meant secrets of the dlc because i read somewhere he said the dlc still has a 'few tricks up their sleeve' although it could be VOG, afterall whats more fun when programming a game than putting easter eggs in. And I would imagine there is more to VOG than is known.

yea idk how you immediately assumed by using the word vault he meant vault of glass, what about the vault space? or vault in the metaphoric sense as to what they might have in store for us.

He may of been using the term "Vault" as a bank. Meaning that they still have some secrets remaining in the game. He didn't specify VoG. I may be wrong but I wouldn't get your hopes up on some new crazy VoG stuff

How is it irrelevant? Vog was included in the how gameplay, but I doubt they'd be adding more stuff to vog this late into the game :/

He was more than likely refering to the treasure room (which they successfully kept hidden until reveal day, no easy feat). And as we know a treasure room is ofcourse a 'vault'. I wouldnt read too much into it.
