I wanna say it's Star Wars, due to the ridiculous amount of canon and content released on an annual basis, but that's one of the few really big ones I'm familiar with. There's just SO MUCH STUFF. You could spend weeks browsing Wookiepedia if you had nothing to do.
Now, this is about sheer vastness and time encompassed, not success or pop-culture presence. For example, Harry Potter is very well-known, but puny in comparison to a universe as defined and expanded-on as the Elder Scrolls.
In fact, TES is noteworthy (in my eyes) because it's very large for such a young franchise. This can be credited to in-game books and the open-world nature, I think.
But what would YOU say is the BIGGEST universe? Remember, non-canon content doesn't count, and neither do crossovers or alternate realities (sorry, comics).

Disregarding everything past the ROTE era, AND not including SWTOR, Star Wars already blimps most science fiction universes out there, [b]Including Warhammer 40K[/b]. If we take all of Star Wars into account, even the BTR era, except for Infinities, it easily lands high on the list.

Warhammer, hands down. The games alone cover two entire eras, there's a -blam!-load of books and the background and fluff is mindblowingly massive.

Lord of the Rings puts Star Wars to shame. I mean, Tolkien freaking developed his own languages and dialects for the different creatures; doesn't get much more developed than that.
