Was your copy of Destiny a [i]digital[/i] copy? If so, then yes if it was purchased before January 15th, 2015.

Yes. Just make sure you're using the same Xbox Live account that purchased the content from your Xbox 360.

Because you have a physical disc, you will have to repurchase Destiny for the Xbox One. Only the digital version of Destiny was applicable for that program. Also, if you redeemed any items via Xbox Live marketplace for Destiny (like this ship, ghost shaders etc) those are platform specific and cannot be transferred as well.
