My theory of the Exo is that they are indeed aliens. Many people seem to assume they are/were built by humans, but for all we know the Exo could be a race of aliens that transcended biological life, becoming 100% machine.
This makes your title question difficult to answer, because we do not really know what the Exo are exactly. If they are cyborgs, than theoretically they can die, because death is the permanent end of all biological function in a living organism, even if that organism has enhanced itself to be primarily or almost completely cybernetic. Now if my theory stands true, and the Exo are sentient machines, then scientifically speaking they can not die, because they are technically not a living organism.
Does this make them expendable? That depends on your definition of life. Technically speaking, a machine is expendable because it can be rebuilt and repaired. But what if this machine can think and act on its own? What if it can experience regret, satisfaction, gratefulness, vengeance and values it's continued existence, just as all biological life does? I don't see why it shouldn't qualify as "living". It's a very hard question to answer, and I probably went on more of a tangent then necessary, but as a final answer I would say no. The Exo are not expendable.

Would there be a need for these makers/inventors of the Exo to imprint themselves at all (there'd certainly be motive)? If sentience can be 100% independent from a biological definition of life, it wouldn't need an imprint or soul to be put into it at all, it could simply manifest itself if given the right conditions, but if those conditions required a machine, then I think death is technically possible, just not requiring biological function, only mechanical. And expendable then becomes an analogous definition to human expendability; more can relatively easily be made, but a unique sentience tied to that function and memory is lost. And if sustaining that uniqueness is valued more than its voluntary or involuntary sacrifice, then it wouldn't be seen as expendable. Expendable is ultimately a value judgement and power relationship; who is doing the spending? -
As I described the Exo I neglected to address the need for a biological imprint, but yes, I would assume it would be necessary. The creators of the Exo more than likely designed them as a conduit to escape the limitations of biological life, while retaining all that they were. If the contents of the brain were digitized, and placed in a new "computer", then there would be a sentience of an organic organism, existing and functioning within a synthetic setting. At this point, all the process going on the Exo's mind are the same process that occur in a biological mind, the exception being where these process are being executed: an organic brain or an electronic processor. If we were to use the modern definition of death, the Exo can not die because they do not have any biological functions. Considering each Exo was once a unique biological organism, each Exo retains the unique sentience of that organism. The retain the desire to keep their unique sentience and attempt to prolong their continued existence as long as possible. In this instance, I would say that expendable is indeed analogous to human expendability. NOTE: Everything in this post is just speculation and opinion. I disagree, it's implied that we are playing the defenders of humanity, holed up in the Last City, setting out into our former realm to reclaim what was lost to us. What reason a transcendent alien race decide to throw their lot in with us? What reason they show up as androids, to make us feel comfortable? The familiarity of their humanoid form chosen to ingratiate themselves to a people who have been driven back to their final refuge by other aliens? It rings false.

I'm not saying that they are aliens, I'm just saying that they might be. We know nothing about the story of Destiny, so really anything goes right now. Maybe the "mind" behind the Exo is an alien creature which has no physical body, and humanity built the Exo to give them a physical form? Or possibly the Exo were designed to protect the species of the universe, and as humanity is on the brink of extinction, they appeared to help us. This is all just speculation, and I tend to let my creativity run on these kind of things.
