My biggest fear is that this "always-online" experience will eventually be plagued by trollers. A lot of the forum community is great, but the forum is a minority.
I fear that in the middle of a firefight I'll be fighting some Cabal on Mars, and then a guy drives up in his pike and throws smoke bombs everywhere to block my vision or drives his pike in front of me, or keeps doing emoticons in my face, or destroys my cover if I'm hiding behind something destructible, or if he has a cool rocket launcher (grenades, grenade launcher) that he keeps shooting at my face so I can't even see anything except smoke and explosions.
A game dependent on a co-op experience with random co-op encounters can easily turn into, (Let me go into the mind of a troll) "Who's game am I going to grief today?" I know it's been confirmed that a guy won't join your game and shoot you in the back, but it's still very easy to troll in different methods.
I'm pretty sure there could be countless other ways to troll (feel free to share/comment), but my biggest fear of Destiny is that this co-op "experience" could easily turn into a co-op nightmare.

My hope is that if that happens, you can Mute that player, like SonOfTheShire stated, and a block from co-op option so you can block that player from coming into your game and messing it up... Bungie wants to have an immersive experience with the best game possible so making sure people can't troll you while you play would be in their top priorities...

They could implement something like the mute feature we have on the forum. If you keep running into a player that proves to be annoying, you could add them to a list of people you don't want to encounter anymore. Something like that would be nice, but it's not like it would really stop it from happening again. I don't know what could be done to prevent it honestly.

To put it simple,nothing is Troll-proof but i'm sure Bungie will implement something to tone it down.

Like 1-50, but geared towards team orientation PVE players, true heroes and offer these players such a game breaking reward that everyone wants it, like Recon or the Haunted Helmet for an example, also it needs to separate these players from the rest so they are being paired up as the top elite of Destiny. With Destiny it shouldn't be about a PVP ranking, K/D, or W/L, but rather who went back to rescue a downed team member or laid down cover fire so someone could make an escape. You're not going to make a troll stop, but if those players who want to have fun in team experiences and positive social efforts can be rewarded and moved up on a social gaming teir acting like a strainer that removes the piss water then all the better for Destiny and the trolling problem down the road. Lastly Bungie needs to really ramp up the monsters and boss battles so it's a team effort with pinning and rescue animations between players and the Ai they face. Trolls existed in Left4Dead but more often than not I'd find players who'd go out of the way to run back to rescue you. Those guys need to be rewarded and not some lip service but something vast that will foster such behavior without hesitation.

I don't think I want something that's troll-proof. I mean yea, I want them to have moderate boundaries, but that's also part of the fun sometimes. Don't get me wrong, kids who make it their sole purpose to ruin a game experience should not be welcome to destroy this new world, but there should be a little room for fun. Some of my best Halo memories are driving teammates off cliffs and hammering a friends warthog into oblivion. I do look forward to seeing how Bungie will make Destiny a positive cooperative experience, but I do hope they leave some room for fun. We have to remember, this is a game :)

I am trying to think of any potential interaction that involves two or more people that is "grief-proof". Can someone provide an example of something that is?

Pretty much what Super Sonic said. You can take measures against trolls but if they want to troll you can't stop them completely.
