To be fair it’s a secret buried in a secret and easy to miss. I did the first part early on but didn’t get the mini-rune. Since then I’ve never gotten symbols on any of my runs to try again.

Fun fact, you don't even have to do the wall symbols to get the rune to spawn. I've done multiple runs and had the rune spawn with or without any other secrets.

Yeah it probably was spawning. All the guides I saw were showing it on the pillar and near the chest, which is where I always looked. Mine was on the wall clear on the other side of the room. Totally random. I could only spot it when I jumped up to a high ledge on the wall to look around. I was on explore when I found it, for those still looking.
