I don't change much of my setup for using these exotics. With the Warlock, I like to max intellect and discipline if I can. I use bloom, so explosions are larger. And I set my melee to charge super faster. in general, I find this exotic more useful for PvP. Obsidian mind would be a better PvE choice, paired with bad juju. Hunter you want maxed discipline, intellect is always a plus. With maxed discipline, by the time one trip mine times out, you will have another. I find this exotic to be effective only in PvP, especially in control. And further for postmortem bounties. Stick it to vertical surfaces on bottlenecks and capture points. Using a weapon with grenadier is good too... Just keep those grenades coming. If you don't have good discipline, you may want to consider just running the symbiote helmet, since you won't be making as good use of the grenades. PvE, both armor pieces aren't as effective. I use the Warlock skull because my raid armor has no intellect, and I prefer void over Sun. Helps me a bunch in PvP bombing control zones.
