Yes they are his true feelings. Deadpool wants, more than anything else, to die. His most wanted wish is to be able to die because he hates his life. Extremely sad for such a comedic and crazy character but it adds so much more depth to him. I forgot the name of the comic but he takes the infinity gauntlet from thanos and holds a massive roast session for all the marvel universe and at the end of it he realizes that no one truly likes him and he purposely made everyone laugh because he wanted the attention and feel accepted. In the second link, which is from that same issue, he calls himself "Pagliacci." Basically reiterating that he makes everyone laugh and happy but they can't make him happy
While I don't know the character enough to say he's depressed and wants to die because of it. I do know that in many of the comics the main reason he wants to die is to be with the woman he loves which is the female form of 'Death' and seeing as he can't die he can't be with her.

Yes it's partly why. He has periodic moments of sanity, which are usually his depressing moments, that he thinks about Vanessa a lot instead.
