The only time you wouldn't use a comma to separate it is when you're using it as a verb or noun, like "HurtfulTurkey's insights always wow me", or "HurtfulTurkey's post was a wow".

Edited by Mad Max: 3/19/2013 11:36:29 PM"Wow" is an exclamation. It is correct to either use it in its own sentence, or separate the exclamation with a comma. "Wow! Look at the tits on that chick!" and "Wow, look at the tits on that chick!" are equally correct.
